The short answer
Yes — a sudoku escape room in a book exists. It is Sudoku with a Secret, Volume 1: Crack the Keeper's Vault by Dan Herlehy: 200 original sudoku puzzles. Hidden among them are keys, each a single shaded cell. Solve the key puzzles and collect their digits. A cipher plate printed in the back of the book converts the digits into letters, spelling three secret words. Each word opens a sealed vault on this website containing free bonus puzzle packs. Nothing is cut or folded, no app is required, and every answer key is printed in the book.
How the secret actually works
Solve
Hidden among the 200 puzzles are grids that carry the Keeper's key mark. Finish one and its shaded square reveals a single digit. Write it on the Vault Door tally page.
Decode
The Keyplate — printed on pages 213 to 215, in the back of the book — turns your digits into letters. The book escalates: the first act is a plain lookup, the second hides its letters in a letter from the Keeper, and the third shifts the alphabet.
Unlock
Each decoded word opens a real vault on this site — 50, 100 and 200 bonus puzzles respectively, as free downloads. No email required. And the Keeper, by his own account, always keeps one secret more.

The closest thing ever published to this book is The Sudoku Code (Heaney & Longo, Sterling, 2006) — long out of print, built on wordoku rather than ciphers, and with no companion site.
